Overview

International name (active ingredient) - mesalazane, 5-aminosalicylic acid, 5-ASA.
Medication form - tablet with delayed release pellets containing mesalazane.

Composition

Each tablet contains a drug Pentas: mesalazane (active substance) 400 mg, povidone, ethylcellulose, magnesium stearate, talc, microcrystalline cellulose -
to 750 mg.

Description

Tablet Pentas is a flat cylinder neravnomernookrashenny yellow-gray, odorless diameter of 13 mm and a height of 4 mm, split groove. On one side of the tablet stamped name of the drug - "Pentasa", on the other - 400 mg.

Pharmacological properties

Pharmacotherapeutic group - anti-inflammatory agent

Pharmacodynamic properties.

Mesalazane inhibits leukocyte chemotaxis, reduces production of cytokinins and leukotrienes, eliminates free radicals.

Pharmacokinetic properties.

The therapeutic effect of mesalazane manifested as a result of local contact of the drug from the intestinal mucosa. After taking a tablet breaks down into micro-pellets acting as independent forms of the drug sustained-release mesalazane. This provides a therapeutic effect of the drug on the way from the duodenum to the rectum at all pH values. Microgranules reach the duodenum during the first hour after ingestion. Passage of the drug on the intact small intestine is performed within 3-4 hours. About 30-50% and 50-80% mesalazane released during passage of the microgranules of the small and large intestine, respectively.
Mesalazane undergoes acetylation in the intestinal mucosa, liver and, to a small extent - enterobacteria. Mesalazane and acetyl-mesalazane not penetrate the blood-brain barrier.
After intravenous injection the half-life mesalazane and acetyl-mesalazane in blood plasma at 40 and 70 minutes respectively.
Mesalazane and its metabolites are excreted in urine and feces.

Indications for Use

Ulcerative colitis (UC) and Crohn's disease of any location.

Contraindications

The drug is contraindicated Pentaca persons with severe liver or kidney function and patients with hypersensitivity to salicylates or other ingredients.

Warnings

Treatment of patients with impaired liver or kidney function is possible only after assessing their severity.
When renal insufficiency Pentas use the drug is not recommended. During the course of treatment, and especially at the beginning should regularly monitor renal function (creatinine level in the blood).
In rare cases, may develop (peri-) myocarditis and pronounced changes in blood counts. If you suspect these side effects should stop treatment and consult a doctor. Manifestations of the above side effects may include: a case of apparent pathological changes in blood - increased bleeding, subcutaneous bleeding, sore throat and fever, or, in the case of (peri-) myocarditis - a fever and chest pain in combination with dyspnea.
In some cases of allergy to sulfasalazine may develop intolerance to the drug and Pentas (risk of allergy to salicylates).

During pregnancy and lactation should appoint a drug Pentaca only in those cases where the positive effect of its use outweigh the risk of possible side effects. Despite the fact that mesalazane crosses the placental barrier, any teratogenic effects on the fetus have been identified. Mesalazane and its metabolite in part excreted in human breast milk feeding women, however, reported any adverse effect of the drug in neonates have been reported.

Not recommended for use Pentaca drug for the treatment of children under the age of 2 years.

The drug Pentas does not affect driving ability.

Dosing and Administration
Pentas is recommended to swallow the pill whole. To facilitate swallowing them can be divided into several parts, or dissolve in water or juice just before the reception.

Side Effects

Typically, drug therapy Pentas well tolerated. However, 1-3% of patients may complain of pain in the abdomen, nausea, vomiting, headache, diarrhea and skin rash in the form of urticaria or eczema.
In rare cases, hypersensitivity reactions to the drug.
By rarely occur side-effects include: muscle aches and joint pain, temporary hair loss, shortness of breath, abnormal liver function and kidney (peri-) myocarditis, pancreatitis and changes in blood counts (leukopenia, granulocytopenia, thrombocytopenia, anemia).
Some of the above expressions can be regarded as symptoms of the disease.

Overdose

Overdoses were observed. Signs of overdose can be expressed by the side effects listed above. Treatment is symptomatic, combined with the monitoring of renal function.

Form release

Each pack contains 10 foil blisters of 10 tablets each.

Pentas tablets should be stored in original packaging at room temperature (15-25 ° C).
